Blood cytology of the common jollytail (Galaxias maculatus) (Jenyns, 1842) (Osmeriformes: Galaxiidae) at postlarval and adult stages

“Puye” (Galaxias maculatus) is a small freshwater fish species of great interest to Chilean aquaculture diversification, because of the high commercial value reached by its transparent larvae or “cristalino”. This study presents the first data with regard to blood cytology in larvae and adults of this specie. Blood smears stained with May Grünwald-Giemsa were analysed through optical microscopy. The results show that in G. maculatus larvae the flowing blood has a transparent look and mature erythrocytes were not observed, as opposed to the adults of this species where the complete erythrocyte developmental line could be observed. In leucocytes, there was no presence of coarse grain cells such as basophils and eosinophyls. In general, the morphology of the blood cells found in larvae and adults of G. maculatus have the same characteristic traits as described elsewhere in reports of other teleosts.
El puye (Galaxias maculatus) es un pequeño pez nativo de gran interés para la diversificación de la acuicultura chilena, debido al alto valor comercial de su estado postlarval cristalino. La presente investigación entrega los primeros antecedentes respecto de la citología sanguínea de postlarvas y adultos de esta especie, determinada mediante microscopía óptica utilizando frotis sanguíneos preparados con May Grünwald-Giemsa. Los resultados muestran que en la sangre circulante de G. maculatus en estado de postlarva no se observan eritrocitos maduros y la sangre es de aspecto transparente. Sin embargo, en adultos se pudo observar la línea eritrocitaria completa y en leucocitos no se observó la presencia de células de grano grueso como son los basófilos y eosinófilos. La morfología de las células encontradas en ambos estados del desarrollo del puye mantienen las características propias entregadas en la literatura para peces teleósteos.

Characterization of a plane on a coordinate measuring machine of large dimensions

A technique for characterizing a horizontal plane within the measuring volume of a coordinate measuring machina (CMM) of large dimensions is described. The objective of the characterization is to obtain a correction table that allows us to minimize the mechanical errors introduced by its mechanical imperfection. The process contemplates the measurement of Y-axis yaw error and the squareness error between the X and Y axes of the machine, as well as obtaining a plane beginning with the characterization of a group of lines. This plane was then used to construct a correction table so that the uncertainty of measurement along the Z axis of the CMM could be reduced to less than 15 micrometers for any measurement carried out within an area of 4X5 m. In addition, a novel method for measuring the deviation angle of a pentaprism using the facilities of the CMM is described.
En éste trabajo se describe un método empleado para caracterizar un plano horizontal dentro del volumen de medición de una máquina de medición por coordenadas de grandes dimensiones, con el propósito de construir una tabla de corrección que permitiera reducir los errores de la máquina introducidos por su imperfección mecánica y de ésta manera reducir la incertidumbre de medición. La descripción incluye el trabajo realizado para obtener el valor del error de deriva del eje Y, el error de perpendicularidad entre los ejes X y Y, así como la obtención del plano a partir de la caracterización de 56 líneas. El plano caracterizado fue empleado para construir una tabla de corrección tal que la incertidumbre de medición en el eje Z se redujo a menos de 15 micrómetros para cualquier punto medido dentro de una área de 4X5 metros. Adicionalmente, se describe también un método novedoso para medir el error en el ángulo de desviación de un pentaprisma y que hace uso de la MMC.

Las cruciferas son una fuente importante de fibra y vitaminas en la dieta del pueblo Colombiano. Su produccion se ve afectada por plagas como Copitarsia sp. que deterioran la cabeza o florete. En este trabajo se estudiaron parametros como fecundidad, fertilidad, mortalidad, proporcion de sexos y ciclo de vida de Copitarsia sp., cuyo conocimiento ayuda al manejo de la plaga. Los estudios se realizaron bajo condiciones naturales de temperatura y humedad relativa en el ICA CA. "Tulio Ospina", uhicado en el municipio de Bello, Antioquia. Los adultos utilizados para estudiar la fecundidad se introdujeron en porrones confiteros que contenian acordeones de papel para la oviposicion. Para el ciclo de vida se partio de una cohorte de 407 huevos, los cuales se mantuvieron individualmente en recipientes plasticos. Cada hembra en promedio puso 550,5 huevos, los cuales tuvieron una fertilidad del 92,87%. La proporcion de sexos fue de 1:1. La mortalidad acumulada en todos los estados de desarrollo fue del 38,82%. El ciclo de vida desde huevo hasta emergencia del adulto duro 45,94 ± 2,5 dias. La incubacion de huevos fue 5,037 ± 0,21 dias. El periodo larval consto de 5 o 6 instares, con duraciones parciales de: LI: 3,80 ± 0,73 dias, L2: 2,80 ± 0,55 dias, L3: 2,97 ± 0,70 dias, L4: 3,12 ± 0,80 dias, L5: 4,91 ± 1,60 y L6: 5,55 ± 0,35 dias; la prepupa y pupa tuvieron duraciones de 2,40 ± 0,62 y 19,26 ± 1,16 dias, respectivamente y el adulto de 15,26 ± 5,37 dias.
The crucifers are an important source of fiber and vitamins in the Colombian diet. Their production is affected by pests like Copitarsia sp., which causes damage to the head or florets of the plant. In this study, parameters like fecundity, fertility, mortality, sex ratio and life cycle of Copitarsia sp. were analyzed because they will support pest management. The study was conducted in ICA "Tulio Ospina" located in Bello, Antioquia under natural conditions of temperature and relative humidity. The adults used to study fecundity were introduced into glass flasks that contained accordion paper used for oviposition. The life cycle began with a cohort of 407 eggs, which were kept in individual plastic containers. Each female lay an average ot 550,5 eggs, which had a fertility of 92,87%. The sex ratio was 1:1 The accumulated mortality in all the developmental stages was 38,82%. The life cycle from egg to adult was 45,94 ± 2,5 days. Egg incubation was 5,037 ± 0,21 days. The larva! period had 5 or 6 instars, with partial durations of: LI 3,80 ± 0,73 days, L2 2,80 ± 0,55 days, L3 2,97 ± 0,70 days, L4 3,12 ± 0,80 days, L5 4,91 ± 1,60 days and L6 5,55 ± 0,35 days; the prepupa and pupa had durations of 2,40 ± 0,62 days and 19,26 ±1,16 days, respectively, and the adult 15,26 ± 5,37 days.

Changes associated with laboratory rearing in antennal sensilla patterns of Triatoma infestans, Rhodnius prolixus, and Rhodnius pallescens (Hemiptera, Reduviidae, Triatominae)

We examined changes in the array of antennal sensilla of three species of Triatominae (Triatoma infestans, Rhodnius prolixus, and R. pallescens) following their establishment for different periods in laboratory culture. In each case, the laboratory colonies were compared with conspecific samples taken directly from the field, by quantitative analysis of the sensilla arrays on the three distal segments of the antenna in terms of the densities of three types of chemoreceptors (basiconics and thick and thin walled trichoids) and one type of mechanoreceptor (bristles). Sensilla densities were compared by ANOVA or non-parametric tests, and by multivariate discriminant analysis. Strains of the same species reared in different laboratories showed significant differences in their sensilla arrays, especially when compared to field-collected material from the same geographic origin. A Bolivian strain of T. infestans reared in the laboratory for 15 years and fed at monthly intervals, showed greatest differences from its conspecific wild forms, especially in terms of reductions in the number of chemoreceptors. By contrast, an Argentine strain of T. infestans reared for 25 years in the laboratory and fed weekly, showed a relative increase in the density of mechanoreceptors. A Colombian strain of R. prolixus reared for 20 years and fed weekly or fortnightly, showed only modest differences in the sensilla array when compared to its wild populations from the same area. However, a Colombian strain of R. pallescens reared for 12 years and fed fortnightly, did show highly significant reductions in one form of chemoreceptor compared to its conspecific wild populations. For all populations, multivariate analysis clearly discriminated between laboratory and field collected specimens, suggesting that artificial rearing can lead to modifications in the sensory array. This not only supports the idea of morphological plasticity in these species, but also suggests caution in the use of long-established laboratory material for experimental studies designed to extrapolate the natural behaviour and physiology of these species.

Susceptibilidad de Plutella xylostella (Lepidoptera: Iponomeutidae) a diferentes aislamientos nativos de Beauveria bassiana y su efecto adicional sobre otros lepidópteros asociados a la coliflor

La polilla dorso de diamante, Plutella xylostella L., es una plaga de importancia en la producción de cruciferas del oriente antioqueño. Se estudió la virulencia de 17 aislamientos nativos y tres productos comerciales de Beauveria bassiana sobre P. xylostella y se seleccionaron los tres con mayor actividad sobre la mencionada plaga; así como el efecto de los tres productos comerciales sobre larvas de Copitarsia sp., Peridroma sp. y Leptophobia aripa (Boisduval), plagas éstas que también están asociadas a la producción de cruciferas en la región mencionada. La patogenicidad del bassiana sobre P. xylostella se caracterizó mediante las variables: porcentaje de mortalidad, tiempo letal medio y mortalidad intrínseca. El efecto en el campo se evaluó en parcelas de 30 plantas, en un diseño de bloques aleatorios, con cinco tratamientos y cuatro repeticiones. Los tratamientos consistieron en dos aislamientos nativos CLS 029 y CLS 003 (que tenían mayor actividad biológica sobre P. xylostella), el producto comercial Bauveril y dos testigos. Las concentraciones letales medias para estos tres tratamientos fueron 2.7xl0(5), 2.1xl0(6) y 2.4xl0(6) c/ml respectivamente. Los tres tratamientos probados causaron mortalidad a P. xylostella en condiciones de campo. Se destaca adicionalmente la infección del aislamiento CLS 029 sobre larvas de L. aripa y del aislamiento CLS 003 sobre Copitarsia sp. Ninguno de los tratamientos afectó a Peridroma sp.
The diamond back moth, Plutella xylostella L., (Lepidoptera: Plutellidae), a pest of signifícance in the production of cruciferas in the Antioquean east. Virulence of 17 native isolations and three commercial producís of B. bassiana on P. xylostella was evaluated. Three of them, wich showed more activity on the mentioned pest, were selected. The effect of these three isolations was also tested on larvae of Copitarsia sp., Peridroma sp. and Leptophobia aripa (Boisduval), which are pests that are also associated with cruciferas in that región. Pathogenicity of B. bassiana on P. xylostella was characterized by means of these variables: percentage of mortality, half-lethal time and intrinsic mortality. The effect in the field was evaluated in plots of 30 plants, in a randomized complete blocks design, with five treatments and four replications. Treatments involve two native isolations CLS 029 and CLS 003 (those with high biological activity on P. xylostella), the commercial product Bauveril and two controls. Half-lethal concentrations for these three, treatments were 2.7xl0(5), 2.1xl0(6) and 2.4xl0(6) c/ml, respectively. The three examined sources of B. bassiana caused mortality to P. xylostella under field conditions. The infection of CLS 029 isolation stands out on L. aripa larvae. Only one larva of Copitarsia sp. was infected with the CLS 003 isolation. None of the treatments affected sp.
